My husband and I will be staying in Montepulciano for 2 nights then rent a car to travel to Florence. This is just a one day rental with drop off in Florence. What would be the best route to visit the what you consider the most beautiful towns along the way? We will have about 9-10 hours available that day.

As for the best route from Montepulciano to Florence I would start by heading west towards Pienza (if you have not already visited it during your 2 days in Montepulciano stop here!), then reach San Quirico d’Orcia but continue north to pass through the scenic Crete Senesi, Buonconvento, Monteroni d’Arbia until you reach the amazing Siena, where I’d suggest you stop for a while, maybe having lunch there.
Then go north for a brief visit to the medieval fortified village of Monteriggioni, then proceed to reach the heart of the Chianti area at Castellina in Chianti. Cross Chianti with any stop you like, and then go north to reach Florence.

Once you get to Monteriggioni you may also run on an alternative route heading west to reach the stunning San Gimignano and its towers. Then, if you still have time drive to reach Chianti, on the contrary, take the fastest way to reach Florence.
