I am planning on hiring a car and driving through Tuscany for 10 days or so. Where should I go [starting from Pisa as I’ve done Florence before] and how many nights in each stop?

Hi! I’m glad to help with your 10 days itinerary in Tuscany Traveling Tuscany by car and for 10 days will give you the opportunity to discover Tuscan destinations that are generally not included among the country’s most popular highlights.
Here is the itinerary I suggest for your vacation in Tuscany:
Day 1: Spend the day visiting Pisa and its marvelous Piazza dei Miracoli, then head toward the walled town of Lucca, simply beautiful, and spend the night there. Lucca has much to offer and is the right place to stay for dinner.
Day 2 and 3: Chianti and San Gimignano: On leaving Lucca head in direction of Chianti; have you already visited it on your first trip? You may still enjoy exploring it and do some wine tastings, while admiring the scenic landscape and the lovely villages atop hills.
Day 4 and 5: Monteriggioni and Siena. The tiny village encircled by massive medieval walls is a great sight! Spend the nights in Siena, that is, in my opinion, one of the most beautiful cities in Italy, enjoy your time there.
Day 6 and 7: Drive south of Siena to reach the Natural Park of the Val D’orcia, incredibly beautiful and with a lot to visit, from stunning hill towns like Montalcino, Bagno Vignoni, San Quirico d’Orcia, and Pienza to suggestive abbeys and unforgettable vistas.
Day 7 and 8: Montepulciano and Cortona. Montepulciano is a perfectly preserved town from the Renaissance, while Cortona, of Etruscan origins, offers an unforgettable panorama.
Day 8, 9, and 10: Orvieto, Civita di Bagnoregio, Pitigliano: Leave Cortona driving south of Tuscany on the border with the Lazio region and visit Orvieto and Civita di Bagnoregio, very impressive and unique hill towns in Italy. Then head toward the suspended village of Pitigliano and the so called villages of tufa: the splendid Sorano and Sovana.
